The Doctor Who star was forced out of most of the London run of the successful Royal Shakespeare Company (RSC) production by a back injury after wowing audiences in Stratford-upon-Avon.
Veteran performers Patrick Stewart and Oliver Ford Davies were nominated in the best supporting actor category for, respectively, the roles of Claudius and Polonius.
The Oliviers recognise excellence on the London stage, so Tennant's injury effectively put him out of the running for the awards.
The nominations see two other seasoned performers up for the best actor prize: Michael Gambon for No Man's Land and Derek Jacobi for Twelfth Night.
Four productions at the Donmar Warehouse - The Chalk Garden, Ivanov, Piaf and Twelfth Night - won 13 nominations between them.
The 33rd annual Laurence Olivier Awards will take place on Sunday March 8 at the Grosvenor House Hotel in London.
